Objectives Calcitonin gene related peptide (CGRP) has been proposed to contribute the transmission of pain in peripheral and central neurons. Mandibular condyle of SAMP8 mice is early‐onset age‐related degenerative changes in TMJ's articular surface (Chen et al., 1989). Osteopontin effect the anchoring osteoclasts in bone remodeling regions. Osteocalcin is also a marker for the bone remodeling produced by osteoblasts. Our study is to identify the expression change of mRNA in CGRP, Osteopontin and Osteocalcin for senescence accelerated mouse model. Methods The senescence‐accelerated SAMP8 mouse (male, 12 weeks of age n=4, 24 weeks of age n=4) and SAMR1 (male, 12 weeks of age n=4, 24 weeks of age n=4) were purchased for this study. All sampled tissues were flash frozen in liquid nitrogen and put in‐80 degree freezer for RNA extraction. Results Quantitative Real time PCR was performed using an Applied Biosystems 7300 Fast Real Time PCR Systems for the detection of CGRP, Osteocalcin and Calcitonin. Our data gave an information for mandible bone and cartilage degeneration accompanied by CGRP expression levels between 12 weeks and 24 weeks. The statistical significance of differences in the percentages of positive cells was analyzed by two‐tailed, unpaired Student's t test ( p <0.01). Conclusion These different expression of each marker also may indicate pain mechanism related to CGRP in mandibular bone. Support or Funding Information none
