Insights into the cellular trafficking of perilipin 5

Perilipins are a family of proteins found on lipid storage droplets that play a role in the regulation and metabolism of cellular neutral lipid. Perilipin 5 has also been found localized elsewhere in the cell including the cytoplasm, mitochondria, endoplasmic reticulum, and more recently, the nucleus. The localization of perilipin 5 in these locations is not random, but regulated in part through phosphorylation of perilipin 5. We hypothesized that the movement of perilipin 5 is regulated through interactions with trafficking proteins. This work seeks to identify the mechanisms and interaction partners involved in phosphorylation dependent perilipin 5 movement in the cell. Following stimulation of the PKA signaling pathway with forskolin and isobutylmethyl xanthine, we observed perilipin 5 localization in the nucleus. The movement of perilipin 5 to the nucleus is time dependent taking minutes to hours to occur. We have identified three weak putative, nuclear localization sequences in the perilipin 5 sequence. Based on these observations, interaction partner(s) must aid the movement of the protein in the cell and transit into the nucleus. These partners may include chaperones, Rabs, importins, or other trafficking proteins. Currently, we are investigating potential interactions between Perilipin 5 and Rabs 32 and 19 using immunofluorescent coverslips and western blotting. Collectively these data indicate that the trafficking of perilipin 5 plays an important role in the function of this protein in the cell.
