Evaluation of The Effects of Organic And Aqueous Extracts of Euphorbia Poissonii on The Gene Expression of BRCA1, BRCA2 and TP53 in MCF‐7 Brest Cancer Cell

Euphorbia poissonii (Euphorbiaceae) is a plant native to Northern Nigeria and it is known to possess cytotoxic properties. The possibility of the crude ethanol extract, hexane, ethyl‐acetate, chloroform, butanol and aqueous fractions to modulate the tumor suppressor genes, BRCA1, BRCA2 and TP53 were investigated. MCF‐7 cell lines were treated with crude ethanolic (CEE), hexane (HF), chloroform(CF), ethyl acetate (EF), butanol (BF) and aqueous (AF) fractions of Euphorbia poissonii and the gene expression of BRCA1, BRCA2 and TP53 were measured using semi‐quantitative RT‐PCR analysis. Results showed down‐regulation of BRCA1, up‐regulation of BRCA2 by CEE, BF, EF, AF and TP53 by HF,CF, EF respectively by the 6 panel fractions. The up‐regulation of BRCA2 and TP53 suggests that the fractions contain bioactive compounds that restored the loss of function of BRCA2 and p53 in the human breast cancer cell line, MCF‐7.
