Effects of bisphosphonate and endurance exercise on cortical bone structure and biomechanical properties in ovariectomized rats

Purpose: To investigate the effects of bisphosphonate and endurance exercise on cortical bone structure and biomechanical properties in ovariectomized rats. Methods Female SD (35 week‐old) rats were assigned to seven groups (n=8‐9 for each group) in accordance with ovariectomy (OVX)/sham surgery (SHA),zoledronic acid (ZA) injection and endurance exercise (EXE), which were: Sham, OVX, OVX+EXE, OVX+ZA4, OVX+ZA4+EXE, OVX+ZA20 and OVX+ZA20+EXE groups. Rats in three EXE groups were subjected to a 12‐week treadmill running training regime (16m/min, 60 min/day, and 5 days/week). Rats of ZA groups were subjected to ZA injections (4 & 20, μg/kg).Methods of micro computed tomography (μCT) and three‐point bending testing were performed on animals' femora. Two‐way (ZA X EXE) ANOVA was used for data processing among ovariectomized groups. T‐test was used for comparing the difference between the SHA and OVX group. Results The SAH group and EXE rats were, respectively, lower than OVX group and non‐EXE rats in body weight (p<0.05). In bone biomechanics, ZA4 and ZA20 were higher than non‐ZA rats in yield load, fracture load, ultimate load, and yield load energy (p<0.05). And, EXE rats was higher than non‐EXE rats in yield load (p<0.05). In bone structure, ZA4 and ZA20 rats were higher non‐ZA rats in cortical bone thickness. Conclusion Both exercise and bisphosphonate showed protective effect on bone strength in ovariectomized rats while exercise revealed additional benefit in body weight loss.
