Difference between hemoglobin‐based oxygen carrier and non‐oxygen carrying plasma expander on cardiac function during hemorrhagic shock resuscitation (545.1)

Study aimed to establish the role of acellular hemoglobin (Hb) based oxygen carrier on cardiac function during anemia. Anesthetized Golden Syrian hamsters under mechanical ventilation with air were hemorrhage 30% of the animal’s blood volume followed by equal volume resuscitation with polymerized albumin (pHSA), or polymerized Hb (pHb, Hemopure, OPK Biotech, Boston, MS). Blood oxygen saturation of >90% was maintained throughout experiment. Cardiac function was assessed through the use of a miniaturized conductance catheter inserted into the left ventricle with continuous pressure‐volume readings. All groups had an increase in stroke work, cardiac output, and ejection fraction compared to baseline. The ratio between dP/dtmax and end diastolic volume, an index of contractility, recovered to 94±33% from baseline for the pHSA, and decreased to 55± 13% from baseline for pHb one hour after resuscitation. To further elucidate the difference in contractility, the intrinsic effects of oxygenation will be investigated by the histology of hypoxic tissue using pimonidazole staining.
