Changes of gene expressions in zinc deficiency and diet restriction in rats (1043.8)

We previously reported that both zinc deficiency and diet restriction affect total RNA concentration and expression of Gapdh and Actb in kidney. In this study, we have examined whether zinc deficiency and diet restriction affect Crebbp and Taf5l in kidney. Thirty 3‐week‐old male Sprague‐Dawley rats were assigned into 3 dietary treatment groups of 10 for the four‐week study: Control group (AIN‐93G), Zn‐deficient group (4.5 mg Zn/kg) and pair‐fed group (AIN‐93G). Kidney was separated into cortex and medulla for RNA isolation. Quantitative real time RT‐PCR using TaqMan proves was performed for mRNAs of Crebbp and Taf5l. Difference between groups was statistically analyzed by Fisher's PLSD. P value less than 0.05 was considered significant. Kidney cortex Crebbp per total RNA was not changed by the treatments. Kidney medulla Crebbp per total RNA was significantly increased by zinc deficiency and diet restriction, respectively. Kidney cortex Taf5l per total RNA was significantly increased by zinc deficiency and diet restriction, respectively. Kidney medulla Taf5l per gram tissue was significantly decreased by zinc deficiency and diet restriction, respectively. These results suggest that zinc deficiency and diet restriction differently affect expression of Crebbp and Taf5l in kidney cortex and medulla.
