γ‐Oryzanol inhibits the adipogenesis of adipose‐derived human mesenchymal stem cells

Gamma‐Oryzanol is one of the most abundant micronutrients in rice bran, reported to have hypocholesterolemic, anti‐inflammatory and antioxidant activities in in vitro and animal studies. To determine the effect of γ‐oryzanol on lipid accumulation, human mesenchymal stem cells were treated with various concentrations of γ‐oryzanol. The cells were obtained from ATCC repository (Manassas, VA) and cultivated in basal medium up to 80% of confluence. After confluence, cells were seeded in 24‐well plates and differentiated into adipocytes for 17 d. During the maintenance phase (d 7–17) cells were treated with 0.1 – 1 μM of γ‐oryzanol (Wako, Japan). MTT assay confirmed no toxicity of γ‐oryzanol up to 10 μM. The medium was replaced every 3 days until the end of differentiation (d 17). Cell supernatants were analyzed 3 days after treatments by a HPLC system. It was found that 43%, 29% and 30% of γ‐oryzanol in the media were taken up by the cells treated with 0.1, 0.5 and 1 μM, respectively. γ‐Oryzanol and its metabolites in the cells are being analyzed by a high‐resolution LC‐MS/MS. Oil red O staining indicated significantly decreased (p<0.01) lipid accumulation by 45, 38, and 35% at 0.1, 0.5, and 1 μM of γ‐oryzanol respectively as compared with control.
