Pentadecapeptide BPC 157 and rat osteoarthritis

Rat osteoarthritis (transected anterior cruciate and medial collateral ligament, medial meniscectomy) exhibited large cartilage destruction, still preserved bone (4 weeks), proximal tibia (cartilage and bone) totally destructed (8 and 12 weeks), and severely impaired knee function. This was rescued with stable gastric pentadecapeptide BPC 157, 10 μg, 10 ng (/kg) either ip once daily or in drinking water (0.16 μg; 0.16 ng/ml/12ml/day) till the sacrifice (at week 4, 8, 12). Knee articular (tibial and femoral) surface similar to nonoperative animals at 4 weeks, only few cartilage lesions at 8 and 12 weeks, thus, cartilage and bone preservation confirmed by X ray exam. Functionally, all parameters analysed (walking, leg pressure force, knee mobility (o)) were improved and rat's leg length maintained. Unlike general failure in controls, BPC 157‐rats walk as non‐operated rats, exhibiting better leg pressure force: i.e., BPC 157‐rats (10μg/kg in drinking water) after 4 weeks 18+/−0.4N (operated) and 20+/−0.7N (nonoperated leg) vs. controls 12+/−1.3 N (operated) and 20+/− 0.5 N (non operated leg); after 12 weeks BPC 157‐rats exhibited 19+/− 0,3N (operated) and 20+/−0.6N (nonoperated), controls only 7+/− 1.1N (operated) and 20+/−1.4N (nonoperated).
