Protective effect of D.mari and Carr. Extracts on oxidative stress in Hydrogen Peroxide‐treated Rat Chondrocytes

The aim of this study is to investigate the protective effect of hot water extract from D.mari and Carr. extracts on cell cytotoxicity and effect on matrix regeneration in rat chondrocytes. The cytotoxic effect of D.mari and Carr. extracts was investigated by MTT assay to determine chondrocyte proliferation against oxidative damage induced by 300 μM H 2 O 2 . The treatment of D.mari and Carr. extracts recovered chondrocytes into a near normal level in a dose‐dependent manner from cytotoxicity by 50%. To examine the effect of plants extracts on matrix regeneration, we measured mRNA expression of proteoglycans and MMPs in chondrocytes. In proteoglycans related to matrix anabolism, type I, II collagen, and pro‐collagen X were strongly expressed with treatment of plants extracts, whereas only MMP‐3 in catabolic signs was reduced compared to untreated control. In conclusion, D.mari and Carr. extracts may be effective to protect chondrocytes proliferation from cell cytotoxicity, and matrix regeneration in chondrocytes could be enhanced by increased expression of proteoglycans such as type I, type II collagen, and pro‐collagen X, and reduced expression of MMP‐3. Thus, D.mari and Carr. extracts could be a possible applicant for ameliorating the degenerative arthritis.
