Megalo types of α‐1,6‐glucosaccharide enhance absorption of quercetin glycosides in rats

We investigated the effects of linear α‐1,6‐glucosaccharides on the absorption of quercetin glycosides in rats. The quercetin glycoside mixture (Q3Gn) and quercetin‐3‐ O ‐glucoside (Q3G) were used in situ and in vivo study, respectively. Q3Gn was composed of Q3G with one or up to seven d ‐glucose adducts. Some linear α‐1,6‐glucosaccharides were enzymatically prepared, and separated into a megalo‐isomaltosaccharide‐containing fraction [M‐IM, average degree of polymerization (DP) =8.4–11.0] and an oligo‐isomaltosaccharide‐containing fraction (O‐IM, average DP=3.3–3.6). M‐IM enhanced absorption of the flavonoid glycosides in the in situ study using small intestinal segment of anesthetized rats. Both α‐1,6‐glucosaccharides increased Q3G concentration in the supernatants of the intestinal contents, and such effect was greater in M‐IM. An in vitro study revealed that the M‐IM was effective for Q3G solubilization. Postprandial plasma level of quercetin‐conjugates was significantly higher in the rats fed M‐IM diet for 14 days than in those fed an α‐1,6‐glucosaccharides‐free diet. In conclusion, M‐IM promotes the absorption rate of Q3G and may contribute to health promotion by Q3G.
