Objective To evaluate through cariometry and estereology if vildagliptin causes morphological changes in the endocrine pancreas of Wistar rats. Methods Sixteen male rats were selected and divided into two groups, control and treated, which received respectively physiological and vildagliptin solution through nasogastric intubation for 10 days. After the sacrifice of all animals and pancreas dissection, histological slices were obtained for cariometric and stereological studies. Computarized programs were used to analyse nuclear parameters of pancreatic islet cells, where insulin is secreted: Major Diameter and Minor Diameter. Mann‐Whitney U Test was applied for statistical analysis. Results and Conclusion Vildagliptina causes significant cariometric and stereological alterations in pancreatic islet cells, with nuclear hyperplasia, without hypertrophy, and an increase in metabolism, with tissue hyperplasia.
