Diosgenin administration improve hyperglycemia through the activation of muscular glucose metabolism

The purpose of this study was to investigate whether administration of diosgenin, a steroid structurally similar to DHEA (dehydroepiandrosterone) and contained highly in dioscorea, improved hyperglycemia in T1DM model rats through the enhancement of muscular GLUT4 signaling. The study was conducted in conformance with the FASEB Statement of Principles for the use of animals in research and education. After one week of STZ injection, fasting glucose level was taken from the tail vein and was monitored for 150min after diosgenin or discorea was injected (3mg/kg). The other day, muscle was taken after 150min of diosgenin or discorea injections. Serum DHEA level was significantly increased after 120min of diosgenin or discorea injections, with concomitant and significant decrease in blood glucose level. Moreover, GLUT4 translocation, as well as phosphorylation of Akt and PKC ζ/λ improved significantly with diosgenin or dioscorea administration. However, these effects of diosgenin and dioscorea were blocked by a dehydrotestosterone DHT inhibitor which inhibited binding to androgen receptor. These results suggest that diosgenin‐induced increase of DHEA level may contribute to the improvement of hyperglycemia through muscular GLUT4 signaling pathway activation in type1 diabetes rats.
