Anti‐neuroinflammaiton and antidepressant effects of Schisandrin B in mice

Forced swimming exercise increases neuroinflammation and changes behavior thus taken as a murine depression‐like model. We investigated the effect of Schisandrin‐B (Schi‐B), one of the active components of Schisandra chinesnesis , on changing depressive behavior and neuroinflammation in mice forced to swim. Stress‐induced depression‐like model mice were prepared by a combination of chronic mild stress and forced swimming within one week. Depression‐like mice were gavaged with vehicle, Schi‐B (10 mg/kg) or Sertraline (32 mg/kg) twice a day for another one week. Sickness behavior, anhedonia, and the immobility time during 5 min of forced swimming test in mice were observed. In depression‐like mice, Schi‐B facilitated the recovery from anorexia, prevented anhedonia and decreased immobility time. Serum serotonin, mRNA expression of serotonin receptor and BDNF in Schi‐B‐treated mice were higher than those of vehicle‐treated mice. Schi‐B treatment decreased indoleamine 2, 3 dioxygenase (IDO) expression, microglia activation and inflammatory cytokines, TNF‐α, IL‐1β and IL‐6, in the brain of mice forced to swim. In conclusion, Schi‐B improved depression‐like behavior in mice forced to swim. The anti‐depressive effect of Schi‐B in mice is related to increase serotonin, serotonin receptor, and BDNF while decrease neuroinflammation.
