Legume consumption and bone mass in ovariectomized rats

This study was examined the effect of consuming soybean, mung bean, cowpea, or adzuki bean on bone mineral density (BMD) and bone biomarkers in ovariectomized (OVX) rats. Sprague‐Dawley rats were either sham‐operated (Sham) or surgically ovariectomized., and then fed AIN‐93M diet (OVX) or AIN‐93M containing soybean (OVX‐S), mung bean (OVX‐M), cowpea (OVX‐C), or adzuki beans (OVX‐A) for 12 weeks. BMDs of the right femur, tibia and lumbar spine were measured using DEXA, and bone morphometric parameters by micro‐CT. The BMD and bone volume of the femur in OVX‐S and OVX‐M groups were significantly higher than OVX group. The expressions of TNF‐α and IL‐6 were significantly lower, while the expressions of osteoprotegerin/RANKL ratio were significantly higher in all bean consumed rats than OVX groups. Serum alkaline phosphatase in OVX‐S and OVX‐M groups was significantly lower, and serum osteocalcin was significantly higher in the OVX‐S, OVX‐M, and OVX‐A groups as compared with OVX group. However, bean consumption had no significant effects on IL‐1β expression, and urinary calcium and deoxypridinoline. In conclusion, soybean and mung bean may prevent bone loss in the OVX rats by reducing TNF‐ α and IL‐6, and increasing osteoprotegerin.
