Mitochondrial reactive oxygen species (ROS) are frequently implicated in various disease states and environmental stressors. Mitochondrial ROS production is not a single process, but rather the net result of several processes including multiple sites of production and elimination. Moreover, the topology of ROS production by mitochondria differs by the site(s) involved which may also impact the relative importance of these products and as the antioxidant systems involved with their further metabolism. This research examines the importance of assay conditions when studying mitochondrial ROS production and consumption, and the interplay between these processes. Particular focus is paid to the influence of external factors such as temperature and pH on these processes. This research is supported by the Canada Research Chairs program and the Canada Fund for Innovation .
