RNA polymerase II (Pol II) transcription in metazoans is punctuated by promoter‐proximal pausing, during which the enzyme begins elongation but pauses during synthesis of the first 100 nucleotides of RNA. This pausing of polymerase is increasingly recognized as an important checkpoint in transcriptional regulation of processes that include response to stimuli, development, and cell differentiation. However, many questions remain concerning the mechanisms underlying establishment vs. release of paused Pol II, and the functional consequences of this mode of regulation. I will discuss our progress towards addressing these questions, with a focus on our recent work investigating the dynamics and stability of paused Pol II.
