Associations between common variants in CYP24A1 and risk of obesity in Chinese Hans

Poor vitamin D status could lead to obesity in Human and gene variants may modify vitamin D metabolism. We aimed to investigate associations of variants in CYP24A1 , encoding 24‐hydroxylase, an enzyme involved in vitamin D degradation, with obesity and its related phenotypes in Chinese Han. Eight tag single‐nucleotide polymorphisms (SNPs) in CYP24A1 (rs2245153, rs927651, rs3787554, rs2181874, rs2762941, rs3886163, rs2248461 and rs2296241) were genotyped in a population‐based sample including 474 obese (BMI ≥28kg/m 2 ), 1221 overweight (24 ≤BMI <28kg/m 2 ) and 1515 normal‐weight subjects (BMI <24kg/m 2 ). After adjustment for age, sex, residential region, season of blood collection and physical activity, five SNPs (rs3787554, rs2181874, rs2762941, rs3886163 and rs2296241) showed significant associations with BMI, waist circumference and/or risks of overweight and obesity ( P ≤0.04). After Bonferroni corrections, the associations of rs3886163 A‐allele (OR = 1.17, P =0.0060) and rs2762941 A‐allele (OR = 1.20, P = 0.0017) with risk of overweight remained significant ( P <0.0063, 0.05/8 tests). In conclusion, the variants in CYP24A1 might contribute to risks of obesity and related traits in Chinese Hans.
