Intestinal Effects of Alanyl‐Glutamine Dipeptide‐Supplemented Diet in a Mouse Model of DSS‐Induced Colitis

Enteral and parenteral glutamine administration has cytoprotective effects in animal models of gut injury. We investigated intestinal effects of oral diet supplemented with alanyl‐glutamine dipeptide (AG) in dextran sulfate sodium (DSS)‐induced colitis in mice. Methods C57BL/6 mice were given 3.5% DSS in water and concomitantly pair‐fed iso‐N oral diets without AG (DSS) or with AG supplementation (AG‐DSS). Control mice did not receive DSS or AG. Mucosal injury scores, myeloperoxidase (MPO) activity, TNF‐α and IL‐6 mRNA (qPCR) and GSH/GSSG redox status were determined in jejunum, ileum and colon after 7 days; clinical status was assessed daily. Results Food intake was similar in all groups. DSS‐treated mice had weight loss, bloody diarrhea, and increased small bowel and colonic MPO activity vs controls regardless of diet (P <0.05). The AG‐DSS group exhibited greater histologic injury in both ileum and colon vs the DSS and control groups (P<0.05). Colonic TNF‐α mRNA was increased in the DSS group vs the AG‐DSS and control groups (P<0.05). IL‐6 mRNA and GSH/GSSG redox in jejunum, ileum and colon was similar in all 3 groups. Conclusion Dietary supplementation with AG increased histologic gut injury and decreased colonic TNF‐α in this model of colitis. Supported by NIH grants K24 RR023356 and T32 DK007298.
