Using a combination of global microRNA knockouts together with re‐introduction of individual microRNAs, we have been dissecting microRNA‐regulated pathways in embryonic stem cells and cancer. I will describe our latest findings in how microRNAs regulate the differentiation and de‐differentiation of stem cells. I will also describe recent results on the role of miRNAs in tumor progression using a mouse model of prostate cancer.
