We present herein a comprehensive and flexible mathematical model of the one‐carbon metabolism network. We have studied the dynamic regulation of the OCM network with a special emphasis on the alteration of DNA methylation and/or dUMP methylation. After validation, the 5mTHF uptake, DNA and dUMP methylation were studied by simulating nutritional 5mTHF deficiency and genetic MTHFR defect conditions. Both conditions show distinct effects on the one‐carbon metabolites synthesis. Methylation reactions shown two oscillations of variable amplitude at a steady state for DNA methylation with severe 5mTHF deficiency and dUMP/dTMP ratio reached the steady‐state after 2h compared to 2.5h for 100% 5mTHF level.
