Interaction between the Renin‐Angiotensin System (RAS) and Reactive Oxygen Species (ROS)

This study investigated whether an interaction exists between intrarenal (IR) RAS and ROS in determining cortical and medullary blood perfusions (CBP and MBP). Groups of male Wistar rats (n=6) were fed a normal Na+ (NNa+, 0.3%) or a high Na+ (HNa+, 3.0%) diet for 2 weeks. Rats were anaesthetized (Chloralose/Urethane) and surgically prepared for measurement of MAP, CBP, MBP and IR infusions. Data, mean ±SEM were analysed using Student's t‐test and significance was taken at P<0.05. MAP was 101±5 and 122±9mmHg, CBP was 82±10 and 158±5 perfusion units (P.U) and MBP was 31±8 and 52±4P.U in NNa+ and HNa+ rats respectively. IR infusion of the SOD inhibitor, DETC, had no effect on MAP but decreased CBP by 24%±6 and 17%±8 and MBP by 20%±5 and 33%±4 (all P<0.05) in NNa+ and HNa+ rats respectively. In the presence of Losartan, the DETC mediated decrease in CBP, but not MBP, was blunted by 10% (P<0.05) in the NNa+ rats but in the HNa+ rats, the fall in MBP, but not CBP was blunted by 21% (P<0.05). These data suggest that angiotensin may interact with ROS but to differing degrees in the cortex and medulla.
