A high fat/high fructose diet has negative effects on brain insulin signaling that are improved by cinnamon

We have shown that cinnamon (CN) and polyphenols in the aqueous extracts of CN counteract insulin resistance in vitro, in experimental animals, and in humans. We determined the effects of CN on brain insulin signaling using a high‐fat/high‐fructose diet (HF/HFr) fed to rats as a model of the metabolic syndrome. Four groups of male Wistar rats were fed for 12 wks with 1) HF/HFr to induce insulin resistance, 2) HF/HFr containing 20g CN/kg of diet, 3) control diet, and 4) control diet containing 20g of CN/kg of diet. The HF/HFr diet decreased whole body insulin sensitivity that was reversed by CN. There was increased gene expression for the insulin receptor and insulin receptor substrate 2, detected by PCR, in the hippocampus of rats fed the HF/HFr diet that was decreased insignificantly in the HF/HFr/CN‐fed group. There was a significant increase in phosphatase and tensin homolog in rats fed HF/HFr, which leads to decreased insulin sensitivity, and which was reversed by CN. Amyloid precursor protein and tau were also increased due to the HF/HFr diet, and CN decreased mRNAs, but not significantly. The HF/HFr‐ and HF/HFr/CN‐induced effects in the cortex were similar to those in the hippocampus but usually smaller. In summary, these data demonstrated that a HF/HFr diet led to negative effects in some insulin related genes in the brain that were reversed by feeding cinnamon.
