Voluntary Exercise Prevents Diet‐Induced Obesity

Background Diet induced obesity (DIO) is a significant health concern. Objective Determine if voluntary exercise (Ex) concurrent with a high fat (HF) diet prevents DIO. Methods Male C57Bl/6 littermates (5 weeks) were distributed equally into 4 groups (n=15–18): low fat (LF) sedentary (LF/Sed), LF Ex (LF/Ex), HF sedentary (HF/Sed) and HF Ex (HF/Ex). Mice were individually housed and LF/Ex and HF/Ex cages had a wheel attached to an odometer to record Ex. Results Reported results are mean ± standard deviation (* indicates p < .05 vs. HF/Sed). HF diet resulted in significantly greater body weight, but Ex prevented this effect (LF/Sed = 29.64* ± 2.67g; HF/Sed = 35.66 ± 5.44g; HF/Ex = 29.6* ± 4.05g). Fasting blood glucose (FBG), oral glucose tolerance (OGTT) and hemoglobin A1c were measured in a subset of mice (n=6–9). FBG was elevated in HF/Sed compared to LF/Sed mice, but not HF/Ex (HF/Sed = 143 ± 22; LF/Sed = 106* ± 6 mg/dL; HF/Ex = 124 ± 25 mg/dL). HF diet induced an elevation in OGTT, but Ex protected against this effect as indicated by the area under the curve (HF/Sed = 44862 ± 7544; LF/Sed = 34150* ± 4626; HF/Ex = 30420* ± 7889). Glycosylated hemoglobin was elevated by HF diet, but not reduced by Ex. Ex resulted in a significant increase in soleus muscle to body weight ratio in the Ex groups indicating a training effect. Conclusion Voluntary Ex attenuated DIO and glucose intolerance, but the mechanism remains to be determined.
