Differential regulation of kidney RAS genes by high sodium intake in lean and obese Zucker rats

Renal tissue Renin Angiotensin System (RAS) plays an important role in many diseases including hypertension. We evaluated transcriptional changes between beneficial components AT 2 , ACE2, and Mas R of RAS alongside deleterious components AT 1A/B , ACE1 and renin in kidney cortex (KC) following 2‐wks of 8% high sodium diet (HSD) in lean and obese Zucker rats (LZR and OZR). We hypothesize that RAS contributes to the pathogenesis of hypertension via a differential regulation of beneficial vs deleterious components of renal RAS in response to HSD. The HSD treatment increased the MAP in both the LZR (16 mmHg) and OZR (21 mmHg) the increase being greater in OZR. The expression data as shown in table below revealed that increased relative expression pattern of renin, ACE1, and AT 1B could be contributing towards elevated MAP in HSD fed animals. Further, higher change in MAP of HSD fed OZR seem to be resulting from increased expression of the deleterious component and concomitant decrease in the expression of beneficial components.
