Effect of Aralia Contientalis extracts in rat primary cultured chondrocytes viability and mRNA expressions related cartilage metabolism under oxidative condition

This study was researched on the effect of Aralia Contientalis(AC) extracts on rat chondrocytes viability and mRNA expression related to cartilage synthesis and erosion in oxidative stress. AC fractions were extracted from 80%, 50%, 5% ethanol and hot water. Chondrocytes were harvested 6~10 weeks Spraque‐Dawley rats and all experiments were fulfilled within the passage number 6. At first, cytotoxity of AC fractions was tested by MTT assay, 80% and 50% EtOH fractions were under the concentration of 200ug/ml, 5% EtOH and hot water fractions were under the concentration of 400ug/ml with no cytotoxity. Cell viability and mRNA expression related cartilage metabolism against the oxidative stress were measured after 300uM of hydrogen peroxide(H 2 O 2 ) treated. All extracts of AC were effective on cell viability in oxidative condition in a dose‐dependant manner, among the AC extracts, 80% EtOH fraction of AC was most effective in cell viability. mRNA expression related to cartilage anabolism and catabolism in oxidative circumstance, genes of cartilage synthesis, aggrecan, collagen type I, collagen type II and procollagen X were most expressed in 80% EtOH AC fraction. mRNAs related cartilage catabolism, MMP‐3, MMP‐7 and MMP‐13 were also expressed under the H 2 O 2 treatment. Consequently, AC 80% ethanol extracts were most effective in rat chondrocytes viability and mRNA expression related to cartilage synthesis in oxidative condition.
