Inhibitory Effect of a Mori Cortex Water Extract on Th2 Cytokine Expression and β‐Hexosaminidase Release by IgE Antigen Complex‐Stimulated RBL2H3 Cells

Mori Cortex is an herbal medicine which has been used in oriental medicine as a traditional therapeutic agent of bronchial asthma but the underlying mechanisms involved are not fully understood. We investigated the antiasthma effects of a Mori Cortex water extract (MCWE) on the β‐hexosaminidase release and expression of interleukin (IL‐4), IL‐13 and tumor necrosis factor‐alpha (TNF‐α) mRNA and protein by IgE‐antigen complex‐stimulated RBL2H3 cells. Stimulation with the IgE‐antigen‐complex induced the release of β‐hexosaminidase and expression of IL‐4, IL‐13 and TNF‐α mRNA and protein. MCWE inhibited IL‐4 and IL‐13 cytokine transcription and translation against stimulation with the IgE‐antigen‐complex and MCWE also inhibited the β‐hexosaminidase release. Thus, MCWE might suppress the Th2‐mediated response through the down regulation of the production of Th2 cytokine and inhibition of degranulation.( This work is Supported for Corporate‐affiliated Research Institute of Academic‐Industrial‐Institutional Cooperation Establishment Business.)
