Sprasis crispa mushroom extract affects adipogenesis and lipolysis in 3T3‐L1 cells

Sprasis crispa (SC, cauliflower mushroom) has high substance of ß‐(1,3)‐D‐Glucan which has various physiological effects on living organism. The purpose of this study was to investigate effects of SC water‐soluble extract in 3T3‐L1 cells. The cytotoxicity of SC in 3T3‐L1 preadipocytes were evaluated by MTT assay. SC treatment (0–1000μg/ml in the preadipocytes) did not show toxicity. Preadipocytes were treated with differentiation cocktail (MDI) in the absence or presence of various concentrations of SC (200,400,600μg/ml). Samples were analyzed on 2nd, 5th and the 8th day of MDI treatment in 24hr, 48hr and 72hr of mature adipocyte. SC had significantly inhibited lipid accumulation compared with control in a time‐and dose‐dependent manner by ORO staining in the adipocyte differentiation and in the mature adipocyte. We also observed increased glycerol release in a dose dependent manner. Furthermore, SC reduces the expression of PPARγ, C/EBPβ in the adipocyte differentiation and increases CPT‐1 in the mature adipocyte. These results indicate that SC suppresses adipogenic enzymes and stimulates lipolytic enzyme providing a basis for anti‐obesity effects of SC.
