Green tea extract reduced myocardial injury in ischemia/reperfusion of rat heart

We recently showed that green tea extract (GTE) can improve the cardiac function by the direct effects on the cardiac contractile apparatus. Since reperfusion injury is one of serious problems in cardiac surgery, we created a rat model of ischemia/reperfusion (IR) by left anterior descending coronary ligation (LAD) for 20 min followed by releasing ligation to determine effects of GTE pretreatment on myocardial protection from IR injury. After LAD ligation, several arrhythmias patterns were defined: GTE significantly suppressed the genesis of arrhythmia after ligation, reperfusion only induced arrhythmias at the initial 5 min with or without GTE. At 24 hr after LAD ligation rats increased serum cTnI levels, while GTE significantly reduced cTnI‐released levels by IR. At 3 days after ligation, echocardiogram showed 26% decrease in Ejection Fraction (EF), and 34% reduction in Fractional shortening (FS), while GTE pretreatment had no changes in EF and FS as compared with controls. In addition, single cardiomyocytes freshly isolated from hearts of control, IR rats with or without GTE at 3 days after ligation showed GTE exerting effects on IR‐induced changes of intracellular Ca 2+ and moderating the cellular distribution of contractile, gap junction, and adhesion proteins in cells.
