Anti‐Proliferative Effects of Sera from Rats with Placental Ischemia on Breast Cancer Cells

Although recent epidemiological studies report preeclampsia has long‐term protective effects against breast cancer, the mechanisms underlying these observations remain unknown. Further, it is unclear whether these effects are due to underlying genetic and environmental factors or are sequelae of the altered endocrine milieu and placental ischemia during preeclampsia. To this end, we hypothesized that serum from pregnant rats with placental ischemia‐induced hypertension contains factors that inhibit proliferation of breast cancer cell lines. A variety of breast cancer cell lines and one lung cancer cell line were seeded onto 48 well plates in media with 5% serum from normal pregnant (NP) or reduced uterine perfusion pressure (RUPP) rats. Decreased proliferation, as measured via sulforhodamine B assay, after 24 or 48 hours of treatment with RUPP serum compared to NP serum was found in MBA‐MB‐231 (62.2% ‐ 24h), Hs578T (20.3% ‐ 48h), MCF‐7 (18.5% ‐ 48h) and UMD227 (79.3% ‐ 24h) breast cancer cell lines. Proliferation of the A549 lung carcinoma cell line was not reduced by treatment with RUPP serum. The present data indicate that placental ischemia may play an important and cell specific role in the anticancer effects observed in formerly preeclamptic women. Understanding the mechanisms responsible for these observations may reveal novel therapeutic approaches to cancer prevention and treatment.
