We have carried out mRNA profiling of 20 esophageal squamous cell carcinoma (ESCC) using whole genome DNA microarrays for identification of novel biomarkers. We chose several novel candidates including periostin (POSTN), lumican (LUM), four and a half LIM domains 1 (FHL1), collagen triple helix repeat containing 1 (CTHRC1) and PTRF interacting protein alpha 1 (PPFIA1) to carry out immunohistochemistry‐based validation on a larger sample set using tissue microarrays. Overexpression of periostin was observed in 139/140 (99%) of cases while. lumican was overexpressed in 141/141(100%) of cases and expression of FHL1 was found to be decreased in 132/145 (91%) of cases. These markers could be further tested in sera of patients as candidates for early detection of this cancer.
