Influence of COX‐2 inhibitor on expression of COX‐2, VEGF, Ang‐1 and Ang‐2 gene in human tongue squamous cell line Tca‐8113

Through observing the change of COX‐2, vascular endothelial growth factor, angipoietin‐1 and angiopoietin‐2 gene in human tongue squamous cell carcinoma Tca‐8113 cell line by ihibiting the activity of cox‐2 to investigate the affect and mechanism of the selective COX‐2 inhibitor nimesulide in the biological behavior of human tongue squamous cell carcinoma.Human tongue squamous cell carcinoma Tca‐8113 cell were cultured in vitro and treated with NIM for 48h to measure the expression of COX‐2, VEGF, Ang‐1 and Ang‐2 mRNA in Tca‐8113 cells by the semiquantitative reverse transcription polymerase chain reaction analysis.Human tongue squamous cell carcinoma Tca‐8113 cell were treated with NIM for 48h,the expression of COX‐2 ,VEGF, Ang‐1, Ang‐2 mRNA level between the each treated group and the control group was statistical significance(P<0.01), The expression of COX‐2, VEGF mRNA reduced in a concentration depengent manner r= ‐0.963,0.934 . Human tongue squamous cell carcinoma Tca‐8113 cell were treated with NIM,the expression of COX‐2, VEGF, Ang‐1 ,Ang‐2 mRNA level in the each treated group compared with the control group was reduced, and the reduced in a concentration depengent manner.
