Immunohistochemical and DNA microarray analysis of rat retinal Müller cells

Gene expression and localization of nutrient‐metabolizing enzymes in retinal Müller cells were examined in order to clarify the meaning of fatty acid beta‐oxidation in the retina. Adult rat retinal tissues and isolated Müller cells from the retina were subjected to western blot analysis, immuno‐fluorescent microscopy and immuno‐electron microscopy for mitochondrial fatty acid beta‐oxidation enzymes. Gene expression of fatty acid beta‐oxidation enzymes and related nutrient‐metabolizing genes were analyzed by DNA microarray. Western blot analysis on adult retina and DNA microarray analysis showed that all the fatty acid beta‐oxidation enzyme examined and their gene expressions are present in the retina but small amount about 1/10 of liver tissue. Immuno‐histochemistry at light and electron microscopy showed all these are present only in Müller cells in the retina and isolated Müller cells, in contrast, they are not present in retinal neurons. Mitochondrial fatty acid beta‐oxidation system was shown to be present in retina, and the Müller cells were sole localization of mitochondrial fatty acid beta‐oxidation system in the retina.
