Antioxidant Effects of Ethyl Acetate‐Soluble Fraction of Chrysanthemum coronarium

The present study was performed to purify phase 2 detoxifying enzyme inducer(s) from Chrysanthemum coronarium. Ethyl acetate‐soluble fraction was loaded on silica column and separated into seven subfractions. Among the subfractions of ethyl acetate‐soluble fraction, subfraction 3 was found to induce NQO1 activity the most strongly in murine hepatoma cells. Furthermore, subfraction 3 induced luciferase activities in HepG2‐C8 cells which were generated by transfecting ARE‐luciferase gene, suggesting ARE‐mediated phase 2 detoxifying enzyme induction. We also investigated whether the subfraction had effect on antioxidant/detoxyfying enzymes in whole animal system. We injected hyrogen peroxide and/or subfraction 3 to ICR male mice, and investigated the expression levels of antioxidant enzymes in tissues as well as FRAP assay. In conclusion, chromatographic subfraction 3 of ethyl acetate fraction of Chrysanthemum coronarium showed strong antioxidant potential and merits identification of active component(s).
