Perilipin gene haplotype is associated with body fatness before and after endurance exercise training

Perilipin proteins are known to regulate intracellular lipolysis in adipocytes. A haplotype of two perlipin gene ( PLIN ) single nucleotide polymorphisms (SNPs), 13041A>G and 14995A>T, has been previously associated with obesity risk. We hypothesized that this haplotype associates with body fatness phenotypes before and after a 6‐mo aerobic exercise training program in sedentary older men and women. The major haplotype group ( n = 57), characterized by the dominant allele at both SNPs (A/A), had significantly lower body mass index (BMI) and percentage, trunk, and subcutaneous fat compared to all other haplotypes (collapsed into a single group, n = 44) before the training intervention (all p < 0.05). After training, BMI, percent and trunk fat remained lower in A/A individuals ( p < 0.05), while subcutaneous fat did not differ between haplotype groups at follow‐up ( p > 0.05) owing to a greater training‐induced decrease in subcutaneous fat in non‐carriers of the A/A haplotype (p < 0.05). Maximal O 2 uptake (VO 2 max) relative to kg body mass was not different ( p > 0.05) at baseline but was greater after training in A/A haplotype carriers ( p < 0.05). This association was not found with VO 2 max after training expressed relative to kg lean mass ( p > 0.05). We conclude that the A/A haplotype of these two PLIN SNPs associates with reduced body fatness before and after training.
