Zinc regulation of bone‐specific transcription factor Runx2 and bone marker gene expression within 24 hours in osteoblastic MC3T3‐E1 cells

Runx2, a bone‐specific transcription factor, increases bone formation through the stimulation of bone‐marker gene transcription in osteoblasts. In our previous study, Runx2 protein expression decreased under Zn deficiency at 24 h with the delayed gene/protein expression pattern, which implies the optimal Runx2 expression would be within 24 h. In the present study, we investigated whether Runx2 protein expression is delayed by Zn deficiency within 24 h and whether the delayed Runx2 protein expression decreased bone marker gene transcription level. Osteoblastic MC3T3‐E1 cells were treated with Zn− (1 μM as ZnCl 2 ) & Zn+ (15 μM as ZnCl 2 ) along with a control (normal osteogenic medium) up to 24 h and collected in every 4 h. Runx2 protein expression was delayed in Zn− having the highest expression at 24 h, while at 16h in Zn+. Protein expression of BMP‐2, as upstream regulator of Runx2, protein expression was also delayed in Zn‐, which is consistent to the Runx2 protein expression pattern. Transcription of bone‐related genes (Pro COLI, ALP, OP, OC & PTHR) showed a delaying pattern, as being consistent to Runx2 protein expression pattern. The study results suggested that Zn deficiency delayed Runx2 protein expression which may be regulated by BMP‐2 protein and again the delayed Runx2 protein expression delayed bone marker gene transcription in 24 h in osteoblasts.
