Effect of Zinc‐ligand Complexes on Intracellular Uptake and Distribution in the Urinary System of Male Sprague‐Dawley Rats

Zinc involves in many cellular metabolisms including gene expression, carcinogenesis, and etc. Especially, prostate gland and testis contain high level of zinc, which is associated with genitals related diseases. We carried out this study to examine the zinc concentration in plasma and various organs in Zn‐ligand complexes and ZnSO4 administered rats, and to investigate the mRNA expression level of zinc transporter. SD rats were divided into three groups: ZnSO4, Zn‐Arg and Zn‐Asp. After oral administration of zinc substrates (40mg zinc/kg body weight), blood and organs were collected. The zinc concentration was detected by zinquin, and zinc transporter mRNA expression was measured by semi quantitative RT‐PCR. We observed that plasma zinc concentration was increased after zinc administration. Plasma zinc concentration of Zn‐Arg and Zn‐Asp group was peaked out at 2h after zinc administration. Although intracellular zinc level in prostate gland and testis is relatively lower than in other organs, zinc level of Zn‐Asp group is higher than other two groups. Also, we found that ZnT2 mRNA expression level was higher in Zn‐Asp group than in Zn‐Arg and ZnSO4 group. This study shows Zn‐ligand complexes are effective the uptake and accumulation of zinc in prostate gland and testis. Therefore, zinc‐ligand complex can be used for available therapy of many zinc‐related diseases of the male genitals.
