IL‐15 and IL‐15R‐alpha SNP Associations with Markers of the Metabolic Syndrome and Adaptations to Resistance Training

The aim of this study was to examine associations between SNPs in the IL‐15 and IL‐15Rá genes and phenotypes in muscle, bone, adipose tissue, and markers of the metabolic syndrome at baseline and in response to resistance training (RT). Subjects consisted of a strength cohort (n=448), MRI volumetric cohort (n=432), and a serum cohort (n=342) who had not performed RT in the previous year. Subjects completed 12 weeks of unilateral upper arm RT using their dominant arm as an untrained control. Strength measurements and muscle, subcutaneous fat, and cortical bone volumes, as measured by MRI, were taken pre‐ and post‐12 weeks of RT, with blood samples taken at baseline. Analyses revealed gender‐specific associations with: IL‐15 SNP (rs1589241) and cholesterol, LDL, HOMA, and BMI; IL‐15 SNP (rs1057972) and difference in 1RM strength, BMI, and fasting glucose; IL‐15Rá SNP (rs2296135) and baseline total bone volume and difference in isometric strength; and IL‐15Rá SNP (rs2228059) and serum triglycerides, baseline whole muscle volume, and baseline cortical bone volume. These data identify associations between IL‐15 and markers of the metabolic syndrome that warrant further investigation. However, these data conflict with a previous report of associations between muscle responses to RT and SNPs in the IL‐15Rá gene.
