Proteomic analysis of human astrocytes treated with propofol

Propofol, an intravenous anesthetic in anesthesia and abirritant in intensive care, is believed to have protective effect on stress of neurons. Astrocytes are connected by gap junctions channel. There is a report that propofol inhibits permeability of gap junction. Therefore, we propose that propofol may exert unknown effects on not only neurons but also astrocytes. The clarification of those effects could contribute elucidation of effect and further clinical application of propofol. We performed proteomic analysis of human astrocytes treated with propofol to address the questions above. We treated human astrocytes with 30 mM propofol in intralipid or intralipid alone for 4 hrs. The cells were collected in sample buffer and homogenized. After centrifugation, the supernatants were applied on two‐dimensional polyacrylamide‐gel electrophoresis. After Sypro Ruby stain, the gel was analyzed using Typoon 9410 with PDQuest software. Several spots, which were significantly elevated levels, were excised and digested with trypsin. The digest was applied on liquid chromatography ion trap mass spectrometer.. We found that these spots were identified as cytoskeletal proteins, such as Coffilin and Fascin. However, morphological changes of astrocyte could not be found. This research is funded by a grant of Saitama Medical University.
