Porcine plasma ficolin binds and neutralizes porcine reproductive and respiratory syndrome virus (PRRSV)

Ficolins are collagenous lectins implicated in innate immune defence against various bacteria, and in the clearance of apoptotic cells. Porcine ficolin‐alpha binds to N‐acetylated saccharides on some serotypes of Actinobacillus pleuropneumoniae but has a limited range of bacterial targets. In these studies, the objective was to identify ficolin gene and protein defects that explain susceptibility of pigs to other common infections. Porcine ficolin‐alpha is expressed predominantly in the liver, and secreted at widely variable concentrations in the plasma of young pigs. We tested the hypothesis that porcine ficolin alpha, either purified from pig plasma by GlcNAc affinity, or as a recombinant expressed in eukaryotic cells, binds to surface glycans on PRRSV that is glycosylated in porcine cells. Purified porcine ficolin alpha had a concentration‐dependent ability to neutralize PRRSV infection in Marc‐145 cells. Also, ficolin was shown to bind to immobilized PRRSV after incubation with porcine plasma or purified ficolin, and this was prevented by preincubation with GlcNAc or PRRSV. These studies indicate that porcine ficolin can interact and neutralize a common arterivirus that is a major pathogen in pigs. Supported by Ontario Pork, CIHR, NSERC and OMAFRA.
