Uroplakins are integral components of urothelial plaques crucial to normal genitourinary function. We examined the effect of gene deletion of uroplakins II (UP2) and III (UP3) on mouse bladder function. Male and female wild type (WT) and knockout (KO) mice lacking uroplakin II (UP2 KO) or uroplakin III (UP3 KO) were evaluated by continuous cystometry in conscious, freely moving animals. Differences in urodynamic parameters (including detrusor overactivity, DO) between genders and between groups were assessed by 2 Way ANOVA. UP2 KO males had larger bladder capacity (BC) than all the other groups. Gender differences between male and female WT mice included: micturition volume (MV) and micturition frequency (MF) (malesfemales). Gender differences in UP2 KO mice included: BC and MV (males>females), basal, threshold, micturition, and intermicturition pressures (males females), and MF (males
