The Tn5 transposition system offers an excellent model system for studying the molecular details of transposase ‐ DNA transactions involved in cut and paste DNA transposition. This is due to the extensive genetic, biochemical and structural information available for the Tn5 transposition system. In addition, Tn5 transposase is a member of the retroviral integrase super family which means that studies on Tn5 transposase can be used to understand other important proteins. In the current studies we will focus on transposition steps not previously elucidated by the known molecular structure. These steps will include the process of transposase binding to transposon DNA in the presence of adjacent donor sequences, the DNA cleavage process that releases transposon DNA from donor DNA and the target capture / strand transfer process that integrates the transposon DNA.
