Grapefruit juice improves trabecular microarchitecture and biomechanical properties in orchidectomized and control rats

Effects of feeding grapefruit juice on bone microarchitecture and strength were evaluated in a male rat model of osteoporosis. Three month‐old male Sprague‐Dawley rats were randomly assigned to orchidectomy (ORX) or sham surgery. Three days post‐surgery, rats were assigned to drink distilled water or fresh squeezed Rio Red grapefruit juice (+GFJ) in a 2 x 2 factorial design. Rats were fed a modified AIN‐93M diet for 2 months. At the end of the study, 4 th lumbar vertebra (L 4 ) were scanned using microcomputed tomography to assess trabecular bone. Strength of the L 4 core was evaluated using finite element analysis (FEA). Data were analyzed using SAS with p<0.05 as significant. ORX significantly reduced bone volume fraction (BV/TV), connectivity density (ConnDens), trabecular number (TbN), and increased trabecular separation (TbSp) showing the importance of testosterone for bone quality. Feeding GFJ increased BV/TV, ConnDens, TbN and reduced TbSp. FEA of L 4 cores showed decreased strain, force for compression, size‐independent stiffness, and increased von Mises stresses due to ORX. GFJ tended to increase strain (p<0.07) and significantly increased force for compression and size‐independent stiffness while decreasing von Mises stresses.
