A polymorphism, 1958G>A, in the 5,10‐methylenetetrahydrofolate dehydrogenase (MTHFD1) gene does not affect choline or folate nutriture in young women consuming controlled choline and folate intakes

The MTHFD1 1958 G>A polymorphism is linked to neural tube defects and is a strong predictor of choline deficiency in pre‐menopausal women when choline intake is severely restricted. This study sought to examine the influence of the MTHFD1 1958 G>A polymorphism on choline status in young women consuming choline intakes that approximated the adequate intake (AI; 425 mg/d) and variable folate (135, 400 and/or 800 mcg/d) intakes. Mexican American women (n=43; 9 GG, 21 GA and 13 AA) aged 18–45 years consumed 135 mcg/d as dietary folate equivalents (DFE) for 7 weeks followed by randomization to 400 or 800 mcg DFE/d for 7 weeks. Throughout the study, total choline (350 mg/d) and betaine (110 mg/d) intakes remained unchanged. Plasma concentrations of betaine, choline, glycerophosphocholine, phosphatidylcholine and sphingomyelin were measured via LC‐MS/MS for weeks 0, 7 and 14. A repeated measures 3‐factor ANOVA was used to examine the effect of week, the MTHFD1 variant, folate intake, and their interactions on choline status. At baseline, no differences were detected among the MTHFD1 genotypes for the choline status parameters. The response of the genotypes to folate restriction and treatment was similar (no week x genotype interaction). These data suggest that the MTHFD1 1958 G>A is not an important determinant of choline status when choline intakes approximate the AI even under conditions of folate restriction.
