The recent declines of Percids (Stizostedion lucioperca and S. volgense) in Russian Kuibyshev basin prompted to study the rapid DNA structuring of their populations. Pikeperches, riverperch (Perca fluviatilis) and ruffe (Gymnocephalus cernuus) are the most important species of this reservoir. The genetic variation and rapidly amplified DNA polymorphism (RAPD) between the exploited fish populations, a product of past evolution in terms of genetics relationship is an essential attempt to increase the overall chances that depleted population will regain and persist over the long period of time. Two microsatellites DNA of walleye (S. vitreum) were used as primers (Svi‐4 and Svi‐18) for this study. Each of the DNA markers resolved 4–6 alleles with a size range of 102–124 bp. The RAPD analysis revealed a diversification of DNA polymorphism in different Percid populations. PCR products of S. volgense have rapidly amplified DNA amplification with a size range of 100 to 1200 bp. Most of the dominant DNA fractions of Percids were existed in the size range of 100–600 bp in both of the primers treatments. The DNA amplification of S. lucioperca had a size range of 100–400 bp with 2–3 dominant DNA band. P. fluviatilis had only one dominant DNA band and ruffe had two DNA amplification signals. Rapidly amplified DNA polymorphism of different Percids depends upon the specific size range of the DNA amplifications. S. volgense evolved as a new species in the way of evolution of the Percid populations. The genetic polymorphism of Percids has a significant variation in among populations.
