The goal of this study is to examine the role of apoptosis in the simple eukaryote S. commune . A putative metacaspase has been identified in S. commune using degenerate PCR with primers based on the conserved regions of the sequence of the yeast metacaspase YCA1. A clone containing the entire genomic sequence of the metacaspase has been isolated and sequenced. In addition, the entire putative open reading frame for the metacaspase was constructed using splicing by overlap extension PCR with genomic DNA and a partial cDNA clone as templates. Currently we are working to clone the open reading frame into an expression vector in order to characterize the enzyme further. Clear sequence similarity between the metacaspase in S. commune and the related enzyme in S. cerevisiae suggests that S. commune has a similar apoptotic mechanism.
