In the yeast Saccharomyces cerevisiae , a mutation in pdr16 results in growth inhibition on the medium chain fatty acid 11:1 10. Previously, we isolated six suppressors of this phenotype in the mutant pdr16 strain, which exhibit intermediate to robust growth on 11:1 10. Two suppressors, 111 and 116, were selected for further study. To identify the 2 nd site mutation responsible for suppression of the pdr16 phenotype, we transformed these strains with a genomic library, and screened for reestablishment of growth sensitivity on 11:1 10 in the suppressors. Plasmids responsible for this phenotype reversion were rescued into E.coli , and sequenced to identify the genomic region contained on the plasmid. Subsequently, fragments were subcloned to identify relevant genes. One such gene, SUT2 , has been identified as responsible for suppression in the pdr16 background. The phenotypic effects of SUT2 are described in this report.
