Relationship of overweight with PPARy2, GRL and CNTF polymorphisms in a Dutch Children Cohort

Background: Genetic variation in the peroxisome proliferation‐activated receptor‐y2 (PPARy2), glucocorticoid receptor (GRL) and neurocytokine ciliary neurotrophic factor (CNTF) genes may play an important role in the etiology of obesity. Objective: we examined the effect of the genetic variation of three polymorphisms on early development and overweight in children at 13 years of age. Methods: In a Dutch cohort of 94 children, anthropometric measurements were determined from birth until 13y. At 7, 12 and 13y, also body composition, leptin concentration and 3 polymorphisms were determined. Results: Children's mean BMI at 13y was 13.4±1.1 kg/m2 and 11% were classified as overweight. The overweight children showed a larger increase in body weight between 4–7y and 12–13y. Children with the PPARy2 polymorphism P/P showed higher leptin levels at 7y and 13y, and a lower BMI at 0.5 y. Children with the polymorphisms G/G of the GRL gene and N/M of the CNTF gene showed higher BMI's up to 4y, and larger weight gain up to 2y. Conclusion: The polymorphisms of the PPARy2, GRL and CNTF gene contribute to the development of overweight in this Dutch Children Cohort.
