TGFBI inhibits endothelial angiogenesis stimulated by adipocyte conditioned medium.

Several studies have suggested that the ECM remodelling or environment is tightly related to the adipogenesis and adipose angiogenesis. In the present study, our data showed that TGFBI (transforming Growth Factor‐Beta–Induced) suppressed angiogenesis induced by adipocyte conditioned medium (CM) in vitro and in vivo. Treatment of TGFBI protein (TGFBIp) diminished the migration and tube‐forming capacity of SVEC‐10 endothelial cells cultured in CM. In contrast, TGFBI knockout (KO) mice contained an increased number of vessels and infiltration into matrigel containing CM. By mechanism of action, TGFBI regulated cleavage and activation of focal adhesion protein (FAK) in adipose tissue. The altered FAK cleavage by TGFBI may account for the modulation of endothelial cell function ultimately impacting adipose angiogenesis. Altogether, these findings demonstrate the potential role of TGFBI in controlling endothelial angiogenesis induced by adipocytes.
