Identification and isolation of the pluripotent stem cells from the Bonghan corpuscle

Even though the pluripotent adult stem cells were recently identified in the several organs of the various species, their exact location are not discovered yet. It is important to find the sites which have the stem cells that could be differentiated to various types of cells. In our previous study, we found novel corpuscle‐like structures which are so called Bonghan corpuscles on the organ‐surfaces and they are connected to various organs by threadlike ducts. We hypothesized that Bonghan corpuscles are supplier of the stem cells in the adult hood, and in order to verify this hypothesis, the sliced Bonghan corpuscles were stained with various stem cell marker antibodies. A few cells in the Bonghan‐corpuscle highly expressed stem cell markers. This was verified by a flow cytometry analysis, showing that a few cells expressed stem cell markers, and also by quantitative‐RT‐PCR and Western blot analysis. To isolate pluripotent stem cells, the Bonghan corpuscles were dissociated and cultured for 12 days and the colony‐forming cells were isolated and analyzed by immunocytochemistry. Almost all of the isolated cells showed high level of stem cell marker expression. When these isolated cells were induced to differentiate, they were differentiated into cells of all three germ layers. The newly identified stem cells in the corpuscle should provide the insight of the mechanism of stem cell function and the opportunities to isolate pluripotent stem cells without severe surgery in the adult.
