SS‐16 BN /Mcwi model of HCM exhibits reduced left ventricular function with age

The SS‐16 BN /Mcwi (SSBN16) rat is a consomic rat with introgression of chromosome 16 from the Brown Norway rat onto the genetic background of the SS/Mcwi (Dahl Salt‐Sensitive (SS)). Previous studies from our laboratory found that SSBN16 rats exhibited hypertrophy of the left ventricle (LV) similar to that seen in SS rats, but without hypertension. This effect was observed on both low (0.4%) and high (4.0%) salt diets. The LV's of SSBN16 also showed interstitial fibrosis and myocyte disarray. The SSBN16 is thus a model of hypertrophic cardiomyopathy (HCM). In this study we wanted to determine if LV function in the SSBN16 rats was impaired beyond that of the SS rat. We monitored the LV function in SSBN16 (n=6) and SS (n=6) rats on a low (0.4%) salt diet from the age of 7 to 16 weeks with weekly transthoracic functional echocardiography. At 7 weeks of age there were no differences in heart rate (HR), stroke volume (SV), cardiac output (CO) or cardiac index (CI) between the strains. By 16 weeks of age HR in the SSBN16 was significantly lower than the SS (291.9±12.9 vs. 385.5±28.1 bpm respectively). There was no significant difference in SV between the two strains. Due to the decreased HR, the CO (ml/min) of the SSBN16 rats was reduced from that of the SS rats at 16 weeks of age (212.1± 8.9 vs. 288.9± 48.1 ml/min respectively). The CI (ml −1 ·min −1 ·g) was also lower in the SSBN16 rats at this age (0.605±0.034 vs.870±0.146 in SS rats). These results suggest that at age 16 weeks the nonhypertensive SSBN16 have LV function that is impaired beyond than that of hypertensive SS. Support HL 29587
